I do not purport that I am by any means an expert in the subject but below I have mentioned a few things that I have learnt over my short time building niche stores and I hope that something is of interest to you.
 
One thing I have learnt is not to rush the stores, research your chosen subject. I have chosen to specialise in things I am interested in, that way it is easy to talk about your products that are advertised on your site.  It is always worth making sure your advertising is product based, this will ensure the adverts are of interest to your prospective customers.
 
The one thing to do is have fun, building and playing around with the layout, colour schemes and content.  Please do not think you will make your millions off your first site.  You will learn a lot as you progress, and no doubt will run into lots of articles on SEO, PPC. This can be daunting; the IT world love's the three syllable acronym. So take your time. The golden place every site developer will tell you is to get to the No.1 spot on Google.  This is hard; millions of people are fighting for this spot.  Planning your strategy to optimising your store and site will take you time. Make sure you sign up to Google’s webmaster tools, this is free and provides excellent tools to help you analyse the traffic coming to your site.

When you get into your site building you will also learn a lot about "Links". Links pointing to your site are invaluable if they are appropriate. Google is very sophisticated and as such over the years has learnt that not all links are good. To get top ratings on Google, you could just pay for lots of links pointing to your site and Google would have thought this was an indication of the site's relevance to the search the user was looking up.  As a result Google has been developed and perfected to rate links and it will only count the links as good links on your site if they are relevant to the content and the search the user is requesting information on. 
 
You can register your site with a number of free directories, these will all put links to your site, if you place your registration in the right category and your site is relevant to that category.
